Solubility
250 nM in 1% (v/v) DMSO / aqueous buffers. Pre-dissolve the complete 10 mg (adjust to the weight given on the vial) in 340 µL DMSO to obtain a 50 mM stock solution. DMSO stock solutions are sterile and can be stored at -20°C for at least 6 months. To avoid too many freeze-thaw cycles, we strongly recommend storage of aliquots. Stepwise dilution: Take e.g. 10 µL of the 50 mM stock solution and dilute by adding 40 µL DMSO to obtain a 10 mM DMSO solution. Take e.g. 5 µL of the 10 mM DMSO solution and dilute by adding 1,995 µL DMSO to obtain a 25 µM DMSO solution. Finally, take e.g. 10 µL of the 25 µM DMSO solution and dilute by adding 990 µL dilution buffer to obtain your final assay concentration (250 nM). Please adjust your calibration to your desired range by measurement of a serial dilution. According to internal assays a nanomolar range is feasible.
Application
Calibrator for fluorescent N-Me-Abz-substrates. Applicable for quantification of fluorescence release, derived from quencher cleavage by transglutaminases in isopeptidase assays.
